Abstract:Enzymes are biochemical reactions catalyzing protein structured molecules. Lipases are enzymes that catalyze lipids hydrolysis reversible in animals and plants under normal conditions. Enzymes are obtained from plants, animals and microorganisms. Obtaining lipases from microorganisms is easier than others in terms of production and preference in terms of financial aspect which takes place cheaper than other sources.Especially extracellular microbial lipases are used frequently in biotechnological applications and organic chemistry. In bacterial lipases produced by the genus Pseudomonas have a wider use in the industry. Being active in a wide temperature and pH ranges, substrate specifity and usefullnes of lipases in biotecnological studies has led to researches on them.In this study extracellular lipase enzyme activity was measured in stationary growth pahase of Pseudomonas putida NRRL B-13 strain by changing environmental conditions such as temperature and amount of enzyme. Effect of MgCl2 cofactor was also investigated on enzyme activity. Enzyme source which was taken from supernatant in 4 ml volume showed the highest enzyme activity at 410 nm. In addition, 10 ml of 0.5 M MgCl added samples have showed the higest enzyme activity at 37 ºC and 42 ºC as well.
