DETERMİNATİON OF TOLUEN BİODEGRADATİON BY PSEUDOMONAS PUTİDA NRRL B-13 STRAİN Sümeyya TATLI Master of Science Thesis Cumhuriyet University Graduate School of Natural Sciences Department of Biology Supervisor: Doc. Dr. Musa SARİ 2019,x+37 pages Pollution, which occurs as a result of contemporary life, is one of the most focused but least solvable issues today. Since the last half of the century, the growing industry and industrial breakthroughs behind the developing technology have increased using the consumption of natural energy sources such as petroleum as they bring with them more energy demand. Cleaning the environment from petroleum hydrocarbons, which significantly affects the biological balance due to its pollutant properties, is very important in terms of environment and applied microbiology. Organic pollutants such as toluene remain in the environment for a long time, threatening environmental safety and environmental health. Toluene occurs during the decomposition of gasoline and from other fuels oil. Biodegradation and bioremediation studies of organic pollutants that pose risks in aquatic and terrestrial ecosystems through microorganisms have gained importance in recent years. Pseudomonas bacterial species have been found to have a tolerance to toxic hydrocarbon groups such as toluene and xylene. This study was planned to determine the degradation of toluene, one of the aromatic hydrocarbons known as an important environmental pollutant, by the Pseudomonas putida NRRLl B-13 strain. Within the scope of this study, Pseudomonas putida NRRL B-13 strain was first produced in liquid media with glucose content and growth curve was created by measuring spectrophotometrically. The bacteria produced were cultured in toluene-containing media and the decrease in the amount of toluene due to time and the increase in the amount of bacteria were measured. Then it was seen that the bacteria did not change their reproduction by decreasing the optimum temperature value. By then the cultivation of bacteria liquid medium with and without magnesium chloride were performed. It was seen that toluen degradation rate was increased in magnesium chloride containing medium. Magnesium acts as a cofactor in the reaction that increasing the degradation of toluen. It was determined that calcium chloride did not affect the biological breakdown of toluene in liquid media.
