Abstract:The aim of this study was to evaluate the penetration of the canal sealer into dentin tubules after calcium hydroxide (Ca(OH)2) removed from the root canal using different irrigation techniques by confocal laser scanning microscopy (CLSM). In the study seventy extracted single-rooted human mandibular premolar teeth were used. Root canals were instrumented with ProTaper Next Ni-Ti rotary files up to X4 (.06/40). Samples were randomly divided into 7 different groups of 10 teeth each. After the root canal shaping procedure, root canals were filled with Ca(OH)2 paste except the control group. All specimens were stored in 37ºC at 100% relative humidity for 1 week. Afterwards Ca(OH)2 were removed from the root canals using different irrigation techniques used: conventional irrigation, EndoActivator, EndoVac, passive ultrasonic irrigation (PUI), neodymium:yttrium-aluminum-garnet (Nd:YAG) laser, photon-induced photoacoustic streaming (PIPS) technique. The canals were dried with paper points and obturated by using AH Plus sealer labeled with Rhodamine B with the cold lateral condensation technique. The teeth were horizontally sectioned at 4, 8, and 12 mm levels from the apex and examined under CLSM to determine the penetration depth, penetration area and penetration percentage of the sealer into the dentin tubules. The data were analysed statistically using One-Way Anova, Tukey and Bonferroni tests. Maximum depth, area and percentage values of sealer penetration inside dentin tubules in the middle and coronal regions of all irrigation techniques were viii higher than the apical region. There was no significantly difference between the groups in terms of maximum penetration depth in the apical and coronal regions (p>0,05). The EndoActivator and the control groups were found significantly different than the conventional irrigation in the middle region (p 0,05). The control group were found significantly different than the conventional irrigation, EndoVac, PUI and Nd:YAG in the middle region (p 0,05). In the coronal region, the EndoActivator was found significantly more effective than PUI (p<0,05). Within the limitations of this study, all of the irrigation techniques were found to be sufficient to remove the Ca(OH)2 medicament in the superficial region of the dentin tubules. It was observed that EndoActivator and PIPS techniques effectively removed Ca(OH)2 from the root canals than other experimental groups.
