Abstract:

In this study, the effects of different preparation sizes and different irrigation techniques applied to mandibular incisors infected with Enterococcus faecalis (E. faecalis) on bacterial reduction were examined. In the study, 112 extracted mandibular incisors with a single root and a single oval-shaped canal were used. Teeth were divided into negative control group and 9 experimental groups. 18 teeth that were sterilized but not inoculated with bacteria were used as negative control group. Sterile teeth in the experimental groups were incubated with E. faecalis suspension for 22 days. 4 teeth were processed for SEM analysis to control biofilm formation. After the biofilm formation was observed in the root canals, the first bacterial samples (S1) were taken from the root canals with sterile paper points. The samples were inoculated on blood agar plates and incubated for 24 hours at 37 °C. Colony forming units were counted and calculated in CFU/ml. The teeth were processed according to the final shaping size and irrigation technique determined for the group they were in. After the final irrigation, post-procedure bacterial samples (S2) were taken and calculated in CFU/ml. SEM analysis was performed on three samples selected from each group to confirm the accuracy of bacterial sampling. The data were statistically analyzed with Kolmogorov-Smirnov test, Kruskal-Wallis test, and Wilcoxon test, and the margin of error was accepted as 0.05. It was observed that there was no bacterial growth in the negative control group. In the pairwise comparison of S1 and S2 data, it was observed that the mean S2 value was significantly lower than the S1 value in all experimental groups (P 0,05). No significant difference was found between the groups that were shaped with different apical preparation size according to the CNI, EA, or PUI methods in terms of S1, S2, S1-S2 and percentage of bacterial reduction (P > 0,05). No significant difference was found between the groups in which different preparation sizes were applied by ignoring the irrigation methods in terms of S1, S2, S1-S2 and the percentage of bacterial reduction (P > 0,05). According to the results of this study, preparation sizes and irrigation techniques were not superior to each other in terms of antibacterial effect.
