Özet

Aim: Epilepsy is estimated by the World Health Organization to affect 0.8% of the world’s population. In recent years, an increasing body of evidence has indicated that there is a complex relationship between epilepsy and the immune system. Adenosine receptors can modulate neuronal activity and could be a powerful therapeutic target for regulating epileptic seizures. The aim of this study is to investigate tumor necrosis factor alpha (TNF-α) and interleukin-1 (IL-1β) levels in the brain of rats in the pentylenetetrazole (PTZ) induced epileptic models in rats. Material and Method: In our study, 24 male 220-240 grams Wistar Albino rats were used. Rats was divided into four groups as control (n=6), Pentylenetetrazole (PTZ 35 mg / kg PTZ was injected every other day to each group intraperitoneally. According to Racine scale, animals with 3 seizures at stage 5 and above were considered to be kindled) Adenosine receptor agonist (A1 agonist) (N6-cyclopentyladenosine 0.3 mg/kg i.p), Adenosine receptor antagonist (A1 Antagonist) (8-Cyclopentyl-1,3-dimetylxanthine (CPX) 20mg/kg i.p). The brain tissues of the animals were removed. After the tissues were homogenized, TNF-α and IL-1β levels were measured in these tissues by using ELISA (enzyme-linked immunosorbent assay) method. The statistical evaluation was done with one-way ANOVA and multiple comparisons were determined by the Tukey test. Results: There was significance increase in TNF-α levels in PTZ group compared to control (p<0.05). Therefore, the increase in TNF-α in A1 agonist group was more than PTZ group (p<0.05). On the other hand, there was significance decrease in TNF-α in A1 antagonist compared to PTZ (p<0.05). Moreover, there was significance increase in IL-1β levels in PTZ group compared to control (p<0.05). However, A1 antagonist reduced IL-1β compared to PTZ (p<0.05). Conclusion: It was found that TNF- α was increased by the A1 agonist and decreased by the A1 antagonist. IL-1β is reduced by the A1 antagonist.
