Sistemik mantar infeksiyonlarÝnÝn tedavisinde kullanÝlan amfoterisin B (amB) ve lipozomal amfoterisin B (lip

lkosit (PMN) fonksiyonlarÝ zerine etkisi araßtÝrÝldÝ. SaÛlÝklÝ gnll donrden elde edilen PMN'ler, in-vitro
koßullarda rhG-CSF'n artan konsantrasyonlarÝ ile muamele edildikten sonra Candida albicans CBS 2730 sußu blastosporlar
ÝnÝ fagosite etmesi ve hcre ii ldrmesi gibi fonksiyonlarÝ gzlemlendi. Ükinci aßamada aynÝ donrlerden elde
edilen PMN'ler antifungal ilalarÝn artan konsantrasyonlarÝ ve C. albicans blastosporlarÝyla birlikte inkbe edildi ve
PMN fonksiyonlarÝ gzlemlendi. nc aßamada ise aynÝ donrlerden elde edilen PMN'ler, nceden rhG-CSF'nin
3000 U/ml konsantrasyonuyla muamele edildikten sonra antifungal ilalarÝn artan konsantrasyonlarÝ ve C. albicans
blastosporlarÝ ile birlikte inkbe edildi ve PMN fonksiyonlarÝ gzlemlendi. BulgularÝmÝz, rhG-CSF'nin sistemik mantar
infeksiyonlarÝnÝn tedavisinde antifungal ilalarla birlikte, zellikle ntropenik hastalarda kullanÝldÝÛÝnda yararlÝ
olabileceÛini dßndrmektedir.
In vitro effects of granulocyte-colony stimulating factor in combination with amphotericin B and liposomal
amphotericin B on human neutrophil functions. In this study combination of amphotericin B and liposomal amphotericin
B, which were used in the treatment of systemic fungal infections with rhG-CSF were tested on human polymorphonuclear
leukocyte (PMN) functions. In the first step, PMNs which were taken from three healthy volunteers were treated
with rhG-CSF in-vitro conditions, and then PMNs were tested with phagocyting and intracellular killing capacity of C. albicans
CBS 2730 blastospores. In the second step, PMNs which were taken from the same donors were incubated with antifungal
drugs and blastospores, and PMN functions tested. In the third step, PMNs which were taken from same donors
were treated with rhG-CSF and then incubated with antifungal drugs and blastospores, and PMN functions tested. As a result
we concluded that rhG-CSF could be helpful in the systemic fungal infections when used with antifungal agents, especially
in neutropenic patients.
