Sistemik mantar infeksiyonlar›n›n tedavisinde kullan›lan flukonazol ve ketokonazolün granülosit-koloni stimüle edici faktör (rhG-CSF) ile kombine edilmesinin polimorfonükleer lökosit (PMN) fonksiyonlar› üzerine etkisi
araflt›r›ld›. Sa¤l›kl›, gönüllü üç donörden elde edilen PMN’ler, in vitro koflullarda rhG-CSF ile muamele edildikten sonra Candida albicans CBS 2730 blastosporlar›n› fagosite etme ve hücre içi öldürme gibi fonksiyonlar› aç›s›ndan gözlemlendi. Ayn› donörlerden elde edilen PMN’ler antifungal ilaçlar ve blastosporlarla birlikte inkübe edildi ve PMN fonksiyonlar› gözlemlendi. Üçüncü aflamada yine ayn› donörlerden elde edilen PMN’ler, önceden 3000 U/ml rhG-CSF ile muamele edildikten sonra antifungal ilaçlar ve blastosporlarla birlikte inkübe edilerek PMN fonksiyonlar› gözlemlendi.
Bulgular›m›z, rhG-CSF’nin sistemik mantar infeksiyonlar›n›n tedavisinde antifungal ilaçlarla, özellikle flukonazol ile
birlikte kullan›ld›¤›nda yararl› olabilece¤ini düflündürmektedir
Summary: Effects of combination with granulocyte-colony stimulating factor, fluconazole and ketoconazole on the
function of human neutrophils. In this study combination of fluconazole and ketoconazole which were used in the treatment of systemic fungal infections with rhG-CSF were tested on neutrophil functions. Neutrophils which were taken from
three healthy volunteers were treated with rhG-CSF in-vitro conditions. Then those neutrophils were tested with phagocyting and intracellular killing of Candida albicans CBS 2730 blastospores. Neutrophils which were taken from the same donors were incubated with antifungal drugs and blastospores, and neutrophil functions tested. In the third step, neutrophils
which were taken from same donors were treated with 3000 U/ml of rhG-CSF and then incubated with antifungal drugs
and blastospores, and neutrophil functions tested. As a result, we concluded that rhG-CSF could be helpful in the treatment of systemic fungal infections when used with ketoconazole, and especially fluconazole.
