A new liquid phase microextraction process has been enhanced for the separation and enrichment of Patent blue V (E 131) dyestuff, which is used in food and medicine. In this method, four different deep eutectic solvents (DES) were prepared and tested as extraction solvents for the selective extraction of Patent blue V. As a deep eutectic solvent; Menthol-levulinic acid with a molar ratio of 1:2 was used, respectively. Within the scope of the method, the effect of matrix types on analytical results was observed. In the context of optimization studies using Box-Behnken Design (BBD), the working range of Patent blue V under optimum conditions was found to be 1–500 μg L−1. The limit of detection and the enrichment factor were calculated as 0.33 μg L−1 and 98, respectively. Visible spectrophotometry was implemented for the determination of Patent blue V in drug and food samples and quantitative analytical data was obtained (relative standard deviation 1.2–2.4 %).
