Goji berry (Lycium barbarum L.) is considered as an economically important perennial herb with high nutraceutical properties. The significant demand for this plant led to optimizing development of an effective in vitro regeneration protocol. Three different explants named leaf, nodal segments, and shoot tips were inoculated on Murashige and (MS) Skoog medium enriched with 1-3 mg L-1 Benzylaminopurine (BAP) and 0.25 mg L-1 Indole butyric acid (IBA). Shoot tip explants induced more shoots compared to other explants. The highest shoot numbers from MS medium was 4.33, augmented with 2.0 mg L-1 BAP+0.25 mgL-1 IBA. After the shoots were rooted successfully on MS medium with 0.5 mg L-1 IBA, they were acclimatized. Additionally, contour plots and surface plots were used to assess the data to make the connection between the input and output variables. When 1.0 mg BAP and 0.25 mg L-1 IBA were combined, the optimization model showed fitted values of 86.67% regeneration frequency with 3.63 shoots per explant. The results open a new window to optimize the different input variables for in vitro regeneration of economically important plants.
